I can't speak for the R6 as I am just finding out all the little secrets to squeeze every pony out of them. I can however tell you that they work well on ZX-12Rs and the R1s.
The R1 only uses two aftermarket stacks. The innermost two are replaced while the outer stacks remain stock OEM parts. The idea is to change the airbox resonance increasing air flow through certain rpms. They ususally don't add topend hp but improve midrange performance and in the case of the R1 add 4-5hp mid range. I've seen the results on my '02 R1 dyno sheet. However this could be completely different for an R6 so....nevermind!
Call Graves Motorsports (gravesmoto.com)and speak with them. They are the Yamaha racing experts.

Yes they work (2-3 hp), but unfortunately they are illegal in all 600 supersport race classes to the best of my knowledge.
